I did 63 miles of this route some time ago, and I can tell you starting from Winchester is generally the best option. Always check the forecast for wind direction. The wind normally blows from the wes... read whole comment...

Description:-
The 160 kilometre long South Downs Way follows the old routes and droveways along the chalk escarpment and ridges of the South Downs. The route provides the visitor with the opportunity "to get away from it all" without having to travel too far in this busy part of England. The undulating route provides a wonderful trip for long distance riders as well as walkers. It also provides interesting day trips and short breaks. The route has little or no technical features but there are very steep gradients in places, sometimes with loose surfaces.

Note that the SDW route has been improved and now ends in Eastbourne in a different place to that shown on the map. The new finish is the same as the footpath version, and is at the end of Dukes Drive, on the westernmost end of the sea front.

Also note that there is an alternative signed route that avoids having to cross the A24 dual carriageway by using a bridge at Washington. It adds extra distance, elevation gain and involves a stretch on a road.

An unsigned alternative route goes via Warnford rather than Exton. This is the original course of the SDW. It involves a road climb. It is quicker and is usually used in "South Downs Double" attempts.
